Study finds osteoclasts block meningeal lymphatic routes to enable dural tumour spread

Researchers using mouse models traced osteoclast-lineage cells (tdTomato+, CtskCre) in bone marrow and dura mater tissue, comparing their marker expression to invading tumour cells. Using confocal imaging and single-cell RNA sequencing, they characterized osteoclasts and infiltrating tumour cells (ITCs) for markers including DLX5, SCX, TNMD, RUNX2 and COL1A1, showing these cells intersect with meningeal lymphatic structures during dural metastasis.

Study finds dural cancer metastases hijack bone cells to damage brain lymphatic vessels

Researchers led by Zeng et al., publishing in Nature, report that tumours spreading to the dura—the brain's outer membrane—recruit bone-degrading osteoclast cells. These osteoclasts damage nearby meningeal lymphatic vessels, which normally drain fluid from around the brain, and weaken local immune defences against the tumour.
